Abstract
Proceso de selección del sexo de espermatozoides mamíferos y método de control de calidad de dosis de sémen sexado congelado. Dicho proceso permite producir por centrifugación en gradiente de densidad, en empresas especializadas en la producción y comercialización de sémen congelado, dosis de sémen enriquecidas con espermatozoides portadores del cromosoma X o Y, sin comprometer la capacidad fecundativa, de estos espermatozoides. Dosis de espermatozoides que contienen ambos espermatozoides X e Y pueden ser separadas para producir subpoblaciones de espermatozoides enriquecidas con portadores del cromosoma X o Y, las cuales son sustancialmente puras con relación al espermatozoide deseado y sustancialmente libres el otro tipo de espermatozoide Reivindicación 1: Proceso de selección del sexo de espermatozoides mamíferos partiendo de la obtención del sémen in natura de reproductores según procedimientos de rutina, caracterizado por comprender la siguientes etapas: a) centrifugación en gradiente de densidad compuesto de medio de cultura químicamente definido para preservar la estabilidad e integridad de la membrana de los espermatozoides; b) utilización de temperaturas de 6 a 22sC, que preservan la viabilidad espermática; c) estabilización del pH en la banda fisiológica para cada especie, preferiblemente alrededor de 7,4; d) asociación a la presencia e sustancias que preserven la integridad espermática, utilizando preferiblemente glucosa, que evita la capacitación y reacción acrosómica precoces, especialmente en bovinos; y e) congelación de los espermatozoides luego de centrifugación en gradiente de densidad en diluidores propios para este propósito, tales como el uso de Tris-yema de huevo, para la utilización en la producción in vitro de embriones (PIV) o en la inseminación artificial (IA) convencional (nacimientos preferiblemente del sexo femenino o masculino o la realizadas después de tratamiento hormonal de las hembras para producir la sincronización del cío e/ou múltiples ovulaciones, cosecha de los embriones ( en su mayoría del sexo femenino o masculino) y posterior transferencia para hembras receptoras. Reivindicación 9: Sémen sexado según cualesquiera de las reivindicaciones anteriores, caracterizado por contener subpoblaciones de espermatozoides enriquecidas con portadores del cromosoma X o Y y ser sometido a las siguientes pruebas: control de calidad e las dosis de sémen que contienen espermatozoides centrifugados en gradiente de densidad, neque dicho protocolo de control de la calidad de fertilidad de los espermatozoides sometidos a sexaje es compuesto por: a) evaluación de motilidad y vigor y prueba de término resistencia (TTR); b) coloración específica para evaluar la integridad de la membrana de los espermatozoides; c) producción in vitro de embriones ; y d) inseminación artificial a través del método comercial.
