Abstract

Determination of the presence of antigens or their anti-bodies in unknown samples using a heterological system of antigens or their antibodies, consisting of: (a) contacting the unknown sample with an antigen or antibody coated on a test object to give a layer of antigen or its antibody obtd. from a first animal species; (b) initial incubation of the unknown sample while in contact with the layer; (c) removal of the unknown sample from the test object and the vessel which tonatin a layer of the antigen or its antibody; (d) contacting the layer with a substance to which an indicator or labelling agent is bound, this material consists of the antigen or its antibody, provided that when the layer consists of antigen, the labelled material is also an antigen but obtd. from a second animal species; (e) second incubation of the layer in contact with the labelled material; and (f) examination of the test object and vessel on the presence of labelled material to determine the presence of antigen or its antibody in the unknown sample. The test may be used on all systems which can utilise a 2-phase antigen- or anti-body system such as antigens with two or more reactive sites or systems which can utilise a 2-phase antibody reaction. Pref. use is to measure heptatitis-associated antigen levels in serum. The test largely prevents cross-reactions in immunological procedures, giving more realistic results.
