Abstract

1,199,731. Basic ketones and derivatives. BEECHAM GROUP Ltd. 24 July, 1967 [23 April, 1966], No. 17879/66. Heading C2C. Novel compounds of Formula I and acid addition salts thereof wherein R<SP>1</SP> is H, OH, or alkoxy, Z and Y together represent an oxygen atom, R<SP>2</SP> is H or alkyl and R<SP>3</SP> is H, alkyl, cyclohexyl, phenylalkyl, dialkylaminoalkyl or hydroxyalkyl or R<SP>2</SP> and R<SP>3</SP> together with N represent a pyrrolidino, piperidino, morpholino, piperazino or 4-alkyl- or 4-hydroxyalkyl-piperazino group, are prepared either by reaction of a compound of formula wherein R<SP>4</SP> # R<SP>1</SP> above, with an amine of formula HNR<SP>2</SP>R<SP>3</SP> and formaldehyde or by reaction of the above amine with a compound of formula When R<SP>1</SP> is OH or alkoxy, the corresponding halo compound, where R<SP>1</SP> is halogen, may be hydrolysed or reacted with an alcohol. The above novel compounds are converted into compounds of general Formula I above wherein X is H and Y is H, hydroxy or acyloxy by reduction of the keto group optionally followed by acylation. Also those which contain a secondary amino group may be acylated and those in which R<SP>1</SP> is H may be halogenated to give compounds wherein R<SP>1</SP> is halogen. 1,1 - Diphenyl - 3 - methylbut - 3 - en - 2 - one is isolated from the reaction of 1,1-diphenylbutan- 2-one formaldehyde and dimethylamine hydrochloride in dimethylformamide. Pharmaceutical compositions having anticonvulsant activity and in conventional forms for oral, parenteral or topical administration comprise an above novel compound and a carrier therefor.
