Abstract

1304707 Benzodiazepines F HOFFMANNLA ROCHE & CO AG 30 June 1970 [2 July 1969 17 Nov 1969 15 April 1970 4 May 1970 13 May 1970] 31501/70 Heading C2C Compounds of the general formula (B=CH 2 , CO; R 1,3,4 =H, C 1-7 alkyl; R 2 =H, halogen) are prepared by (a) diazotizing a corresponding 7-amino derivative, treating the diazonium salt with an alkanoyl group-providing agent and hydrolysing the product, (b) ring- closing a compound of the formula (c) treating a corresponding 7-cyano derivative with an alkyl lithium and hydrolysing the product (B = CH 2 when R 1 = alkyl) or (d) oxidizing the α-methylene group in a corresponding 7-(C 1-8 alkyl) derivative, optionally followed in each case by N-alkylation and/or oxidation of B = CH2 to B=CO. The above compounds are anticonvulsants, muscle relaxants and sedatives and may be administered in the form of pharmaceutical preparations containing them, with or without other (unspecified) active ingredients, in association with a carrier.
